The Use of Amantadine in Parkinson’s Disease and other Akinetic-Rigid Disorders

چکیده

Introduction Amantadine was originally introduced as an antiviral agent to treat influenza A and was coincidentally found to ameliorate symptoms in a patient with Parkinson’s Disease (PD) in 1969. Since that time many clinical trials have investigated the efficacy of amantadine alone and in combination with other antiparkinsonian drugs. Most of these trials took place in the early 1970s. More recently, the use of amantadine has focused upon the treatment of levodopa-induced motor fluctuations and dyskinesias. This article will summarise the evidence available for the use of amantadine in PD and other movement disorders.

A prospective study of freezing of gait with early Parkinson disease in Chinese patients

This study investigated the risk factors for freezing of gait (FOG) in the early stage of Parkinson disease in China, using a sample of 248 patients who were followed for 3 years. Part III of the Unified Parkinson Disease Rating Scale and the modified Hoehn-Yahr grading scale were used to evaluate the severity of motor symptoms. Nonmotor symptoms were assessed using the Hamilton Anxiety Rating ...

Amantadine: an antiviral and antiparkinsonian agent

Amantadine is an antiviral agent that specifically inhibits influenza A virus replication at a micromolar concentration. This drug is also very effective in the treatment of human Parkinson’s disease. Other important clinical applications of this agent have been studied recently, ranging from viral infections, e.g. herpes, herpes zoster neuralgia to granulomatosis and from neuroleptic extrapyra...

SHORT REPORT Duration of amantadine benefit on dyskinesia of severe Parkinson’s disease

Background: Recent short-term studies suggested that amantadine (Ama) might ameliorate dyskinesia in patients with Parkinson’s disease. A double-blind study programmed over 12 months was designed to assess the duration of the antidyskinetic effect of amantadine on levodopa induced dyskinesia. Methods: 40 patients treated for 7.5 (2.2) years with levodopa (729.3 (199.4) mg/day) and dopaminoagoni...

Striatal inhomogeneities and basal ganglia function.

It has always been something of a mystery as to why different diseases of the basal ganglia may produce completely opposite effects on movement. Thus, defective dopaminergic inhibitory input into the striatum in Parkinson’s disease causes an akinetic-rigid syndrome, while destruction of the striatum in Huntington’s disease usually causes chorea. This theoretic paradox may be resolved by more de...

What contributes to quality of life in patients with Parkinson’s disease?

Objective—To identify the factors that determine quality of life (QoL) in patients with idiopathic Parkinson’s disease in a population based sample. Quality of life (QoL) is increasingly recognised as a critical measure in health care as it incorporates the patients’ own perspective of their health. Methods—All patients with Parkinson’s disease seen in a population based study on the prevalence...
